Protein production and crystallization
We offer efficient, high-throughput production of high purity proteins for crystallization and structure solution. Working with a variety of protein expression vectors and batches of expressed proteins we are able to explore the ideal route to a protein structure.
Protein production
Up to 96 protein expression constructs can be prepared for a single target in one batch to rapidly explore the optimum construct length, and possible affinity purification tags. Constructs are tested in small-scale expression in E.coli or insect cells to identify the optimum expression methodologies and scaled up for purification.
Protein purification can be performed on multiple proteins in automated, overnight experiments, offering maximum efficiency and rapid turnaround of a full range of protein variants. We commonly achieve a protein purity of at least 95%.
Protein crystallization
We carry out initial crystallization trials using automated, sub-milligram 96-well screens, reducing the time and cost of producing and purifying larger quantities of protein. Proteins are screened with a range of known ligands in order to identify the best conditions for crystal growth. Initial success is followed by further automated optimization to refine the crystallization process.
Our expertise means we can crystallize liganded proteins, either through co-crystallization or by soaking of ligands into pre-formed crystals.
